Title:  
A Phase II Trial of Autologous Stem Cell Transplant Followed by Miniallogeneic Stem Cell Transplant in Lieu of Standard Allogeneic Bone Marrow Transplantation for Treatment of Multiple Myeloma
Phase:  
II
Purpose:  
To determine the effectiveness of receiving an allogeneic transplant after an autologous transplant to treat myeloma
Eligibility:  
  • Diagnosis of myeloma
  • 18 to 70 years old
  • No previous autologous or allogeneic transplant
  • adequate heart, liver, kidney function
  • No nonsecretory myeloma
  • HLA-matched sibling is needed
Treatment:  
Patients receive an IV injection of melphalan the day before receiving an autologous stem cell transplant. The day after the stem cells have been harvested and reinfused, patients receive G-CSF, either by injection or IV until white blood cell counts have recovered and are stable for 3 days. Approximately 100 to 182 days after patient's own stem cells are reinfused, the miniallogeneic procedure begins including fludarabine and cyclophosphamide over 5 days followed by and donor stem cell infusion. A combination of cyclosporine and solumedrol are administered for up to 4 months after the miniallogeneic transplant to help with acceptance of the new cells and to help prevent graft vs host disease
